Myosin light chain kinase (MLCK) is a serine/threonine kinase that belongs to CaM Kinases (Ca++/Calmodulin-dependent protein kinases) family. MLCK is involved in the regulation of myosin activity during smooth muscle contraction. At increased level of Ca2+, rapid binding of Ca2+ to Calmodulin (CaM) occurs, triggering MLCK activation.

Myosin light-chain kinase also known as MYLK or MLCK is a serine/threonine-specific protein kinase that phosphorylates a specific myosin light chain, namely, the regulatory light chain of myosin II. [3]

2 Physiologie Myosin light chain kinase (MLCK) is a Ca 2+ –CaM-dependent enzyme that phosphorylates MLC at Ser-19 or Ser-19/Thr-18 (Dudek and Garcia, 2001; Lazar and Garcia, 1999; Verin et al., 1998a,b; Wadgaonkar et al., 2003). Previous biochemical evidence shows myosin light chain kinase binds tightly to actomyosin containing filaments. The kinase has low-affinity myosin and actin binding sites in Ig-like motifs at the N- and C-terminus, respectively. Recent results show the N-terminus of myosin light chain kinase is responsible for filament binding in vivo.

p21-activated kinases (PAKs) are implicated in the cytoskeletal changes induced by the Rho family of guanosine triphosphatases. Cytoskeletal dynamics are primarily modulated by interactions of actin and myosin II that are regulated by myosin light chain kinase (MLCK)–mediated phosphorylation of the regulatory myosin light chain (MLC). p21-activated kinase 1 (PAK1) phosphorylates MLCK
